Product Description
EGFP mRNA serves as a reporter mRNA, making it an ideal choice for monitoring and optimizing transfection efficiency; it can be utilized as a positive control for mRNA transfection experiments. This mRNA was synthesized using T7 RNA polymerase, utilizing a linear double-stranded DNA template containing a T7 promoter sequence and NTPs as substrates. All UTPs were replaced with N1-Me-PseudoUTP, and Cap1 was added to enable simultaneous capping during the in vitro transcription process, thereby yielding EGFP mRNA with a high capping efficiency. Compared to conventional UTP, modification with N1-Me-PseudoUTP results in higher translational efficiency and suppresses mRNA-mediated innate immune activation.
Experimental Case Studies
Using the Lipofectamine 2000 transfection reagent, 1 µg of EGFP mRNA was transfected into HEK 293T cells. Twenty hours post-transfection, significant expression of EGFP was observed via fluorescence microscopy.
